Paradox: Curcumin, a Natural Antioxidant, Suppresses Osteosarcoma Cells via Excessive Reactive Oxygen Species

Osteosarcoma (OS) is an aggressive tumor with a rare incidence. Extended surgical resections are the prevalent treatment for OS, which may cause critical-size bone defects. These defects lead to dysfunction, weakening post-surgical quality of patients’ life. Hence, ideal therapeutic agent OS should simultaneously possess anti-cancer and repair capacities. Curcumin (CUR) has been reported in therapy regeneration. However, it not clear how CUR suppresses development. Conventionally, considered natural antioxidant line its capacity promote nuclear translocation transcription factor, factor erythroid 2 (NRF2). After translocation, NRF2 can activate some antioxidases, thereby circumventing excess reactive oxygen species (ROS) that deleterious cells. Intriguingly, this research demonstrated that, vitro, 10 20 μM increased intracellular ROS MG-63 cells, damaged cells’ DNA, finally caused apoptosis although protein level expression NRF2-regulated antioxidase genes were identified those two groups.
